Formation Of Nucleus Pulposus-Induced Disc Hernia-Like Nodules On The Disc Surface Is Not Induced By TNF
نویسنده
چکیده
It was recently seen that puncture of an intervertebral disc in the rat induced disc hernia-like nodules over the puncture site and that the presence of nucleus pulposus was necessary to produce such nodules. Since TNF from the nucleus pulposus have been found to be involved in pain production the aim was to see if also TNF was involved in the formation of such nodules. Two TNF-inhibitors, infliximab and methotrexate, were administered to rats following disc puncture. Non-treated animals were used as controls. Macroscopic evaluation after one and two weeks did not demonstrate any notable differences between non-treated and treated animals. The data thus indicate that TNF is not involved in the formation of nucleus pulposus-induced disc hernialike nodules on the disc surface. This may instead be induced by other bioactive substances derived from the nucleus pulposus, such as growth factors. However, further studies with direct administration of TNF-inhibitors must also be performed to understand the contribution of TNF in this regard. CONFLICT OF INTEREST The author is CEO and part owner of the company Sciaticon AB that holds patent on pharmacological treatment of low back pain and sciatica by TNF-inhibition.

Puncture of a Disc and Application of Nucleus Pulposus Induces Disc Herniation-Like Changes and Osteophytes. An Experimental Study in Rats
It has been observed that puncture of a lumbar disc may induce formation of a nodule on the surface of the disc and osteophytes. It is not known if this is based on the presence of a foreign tissue or specifically by the presence of nucleus pulposus or on the disc injury. In this study these mechanisms were separated by comparing disc puncture with application of nucleus pulposus without disc i...
متن کاملGallic acid inhibits the release of ADAMTS4 in nucleus pulposus cells by inhibiting p65 phosphorylation and acetylation of the NF-κB signaling pathway
This study investigated the inhibitory effect of gallic acid (GA) on the release of A Disintegrin and Metalloproteinase with Thrombospondin motifs 4 (ADAMTS4) through the regulation of the NF-κB signaling pathway, which is closely related to the matrix metalloproteinases in nucleus pulposus cells. Different concentrations of GA were added to TNF-α-induced human nucleus pulposus cells (hNPCs) an...
متن کاملEffects of Morphine Dependency on Intervertebral Disc in the Rat
Background: A large proportion of patients presenting with lumbar disc disease are opium addicts in our region. It was, therefore, hypothesized that morphine might affect the intervertebral disc. We investigated the histological findings of morphine dependency on intervertebral discs in rat. Methods: Forty NMRI adult male rats (230-250 g) placed on ordinary diet and received aqueous solution of...

Objective(s): Activation of acid-sensing ion channel 1a (ASIC1a) is responsible for tissue injury caused by acidosis in nervous systems. But its physiological and pathological roles in nucleus pulposus cells (NPCs) are unclear. The aim of this study is to investigate whether ASIC1a regulates the survival of NPCs in the acidic environment of degenerated discs. Materials and Methods: NPCs were i...
متن کاملFollistatin-like protein 1 promotes inflammatory reactions in nucleus pulposus cells by interacting with the MAPK and NFκB signaling pathways
OBJECTIVE Follistatin-like protein 1 (FSTL1) is a well-known mediator of inflammation. Intervertebral disc disease is an inflammatory disorder. Here, we investigated the role of FSTL1 in the intervertebral discs inflammation. METHODS Expression of FSTL1 in nucleus pulposus tissues from rats and human was determined by immunohistochemistry staining and western blot analysis. The expression lev...
